Newly identified natural protein blocks HIV, other viruses

Feb. 11, 2013 ? A team of UCLA-led researchers has identified a protein with broad virus-fighting properties that potentially could be used as a weapon against deadly human pathogenic viruses such as HIV, Ebola, Rift Valley Fever, Nipah and others designated "priority pathogens" for national biosecurity purposes by the National Institute of Allergy and Infectious Disease.

In a study published in the January issue of the journal Immunity, the researchers describe the novel antiviral property of the protein, cholesterol-25-hydroxylase (CH25H), an enzyme that converts cholesterol to an oxysterol called 25-hydroxycholesterol (25HC), which can permeate a cell's wall and block a virus from getting in.

Interestingly, the CH25H enzyme is activated by interferon, an essential antiviral cell-signaling protein produced in the body, said lead author Su-Yang Liu, a student in the department of microbiology, immunology and molecular genetics at the David Geffen School of Medicine at UCLA.

"Antiviral genes have been hard to apply for therapeutic purposes because it is difficult to express genes in cells," said Liu, who performed the study with principal investigator Genhong Cheng, a professor of microbiology, immunology and molecular genetics. "CH25H, however, produces a natural, soluble oxysterol that can be synthesized and administered.

"Also, our initial studies showing that 25HC can inhibit HIV growth in vivo should prompt further study into membrane-modifying cholesterols that inhibit viruses," he added.

The discovery is particularly relevant to efforts to develop broad-spectrum antivirals against an increasing number of emerging viral pathogens, Liu said.

Working with Jerome Zack, a professor of microbiology, immunology and molecular genetics and an associate director of the UCLA AIDS Institute, the researchers initially found that 25HC dramatically inhibited HIV in cell cultures. Next, they administered 25HC in mice implanted with human tissues and found that it significantly reduced their HIV load within seven days. The 25HC also reversed the T-cell depletion caused by HIV.

By contrast, mice that had the CH25H gene knocked out were more susceptible to a mouse gammaherpes virus, the researchers found.

In collaboration with Dr. Benhur Lee, a professor of pathology and laboratory medicine and a member of the UCLA AIDS Institute, they discovered that 25HC inhibited HIV entry into the cell. Furthermore, in cell cultures, it was found to inhibit the growth of other deadly viruses, such as Ebola, Nipah and the Rift Valley Fever virus.

Intriguingly, CH25H expression in cells requires interferon. While interferon has been known for more than 60 years to be a critical part of the body's natural defense mechanism against viruses, the protein itself does not have any antiviral properties. Rather, it triggers the expression of many antiviral genes. While other studies have identified some antiviral genes that are activated by interferon, this research gives the first description of an interferon-induced antiviral oxysterol through the activation of the enzyme CH25H. It provides a link to how interferon can cause inhibition of viral membrane fusion, Liu said.

He noted some weaknesses in the research. For instance, 25HC is difficult to deliver in large doses, and its antiviral effect against Ebola, Nipah and other highly pathogenic viruses have yet to be tested in vivo. Also, the researchers still need to compare 25HC's antiviral effect against other HIV antivirals.

Additional study co-authors were Roghiyh Aliyari, Kelechi Chikere, Matthew D. Marsden and Olivier Pernet, of UCLA; Jennifer K. Smith, Rebecca Nusbaum and Alexander N. Frieberg, of the University of Texas-Galveston; and Guangming Li, Haitao Guo and Lishan Su, of the University of North Carolina-Chapel Hill.

Supernova Alert! Astronomers Spot Warning Outburst

Forecasting when stars will die in giant explosions may one day be possible by looking for the warning outbursts they release beforehand, researchers say.

Supernovas are the most powerful stellar explosions in the universe, visible all the way to the edge of the cosmos. These stars detonate for two known reasons: either from gorging on too much mass stolen from a companion star or by running out of fuel and abruptly collapsing.

Astronomers have suggested that stars can give off smaller explosions just before they go supernova. To find out more about supernovas, researchers used three telescopes ? the Palomar Observatory, the Very Large Array and NASA's Swift mission ? to investigate a star 500 million light-years away. The star, which had about 50 times the mass of the sun, ultimately detonated as a supernova named SN 2010mc.

The researchers' data suggest that 40 days before the final explosion, the dying star produced a giant outburst, releasing as much matter as 1 percent the mass of the sun ? about 3,330 times the mass of Earth ? at about 4.5 million mph (7.2 million km/h). [Photos of Great Supernova Explosions]

"What is surprising is the short time between the precursor eruption and the eventual supernova explosion; one month is an extremely tiny fraction of the 10-million-year lifespan of a star," said one of the study authors, Mansi Kasliwal at the Carnegie Institution for Science in Pasadena, Calif.

This explosion radiated "about a million times more than the energy output of the sun in an entire year," author Mark Sullivan of the University of Southampton in England told SPACE.com. But this precursor "is still about 5,000 times less than the energy output of the subsequent supernova."

The close timing between the outburst and the ensuing supernova suggest they are related, lead author Eran Ofek of the Weizmann Institute of Science in Israel said in an email interview. Probability models revealed there was only a 0.1 percent chance that the outburst was a random event.

"Our discovery of SN 2010mc shows that we can mark the imminent death of a massive star. By predicting the explosion, we can catch it in the act," Kasliwal said.

Comparing their data with three models proposed for how the preceding explosion might have occurred, the researchers found that gravity waves helped drive mass to the star's atmosphere. Gravity waves are fluctuations caused by matter rising due to buoyancy and sinking due to gravity.

"For a star like our sun, the energy it is emitting from the fusion of hydrogen into helium deep in the core exerts an outward pressure on the star, usually counteracted by an inward pressure from gravity. However, if the star's luminosity increases above a certain amount ? the so-called Eddington luminosity ? the outward pressure from the resulting radiation is strong enough to overcome the gravity, which can then power an outflow of material," Sullivan explained. "Gravity waves can act as a conduit to translate this large, super-Eddington luminosity in the core into an ejection of material from the outer envelope of the star, just like we observed."

The scientists detail their findings in the Feb. 7 issue of the journal Nature.

McDonald's sales fall with tough year ahead

(AP) ? McDonald's says a key sales figure dropped again in January as the world's biggest hamburger chain struggled to grow in the U.S. and saw a severe decline in Asia.

The Oak Brook, Ill.-based company said global sales at restaurants open at least 13 months dropped 1.9 percent for the month. The figure is a key metric because it strips out the volatility of newly opened and closed locations.

After years of outperforming rivals, McDonald's has been struggling amid intensifying competition and challenging economic conditions around the world. Late last year, the company ousted the head of its U.S. business after the sales figure dropped for the first time in nearly a decade. CEO Don Thompson, who took the top spot this summer, has vowed to add business by emphasizing value while planning a series of new limited-time offers to attract customers.

But Thompson also warned last month that sales were trending negative for January. The company was also up against a tough comparison from a year ago, when global sales rose 6.7 percent.

In the region encompassing Asia, the Middle East and Africa, McDonald's said its sales sank 9.5 percent in January. The company is facing particular difficulty in Japan, where it says customers have been eating at home more often since the earthquake and tsunami in 2011. It also recently shifted its marketing strategy to focus on staples like the Big Mac and breakfast, rather than promotional items such as the "Big America Series" from a year ago. The sales figure fell 17 percent as a result.

In China, the company said the timing of Chinese New Year hurt results. But another factor was the ongoing wariness among diners after reports on Chinese television that chicken producers were ignoring regulators and giving birds unapproved levels of antibiotics. Yum Brands, which owns KFC and is the biggest Western chain the country, has been slammed by the reports and expects its sales in the country to plummet by 25 percent in the first quarter.

In Europe, McDonald's biggest market, sales fell 2.1 percent as Germany and France dragged down results. The company said it remains focused on enhanced value and keeping stores open longer in the region.

In the U.S., McDonald's said the figure edged up 0.9 percent. The addition of the Grilled Onion Cheddar burger to the Dollar Menu boosted results.

For February, McDonald's noted sales will be hurt by about 3 percentage points since the previous year's results included an extra day because of the leap year.

The monthly sales figures are a snapshot of money spent on food at both company-owned and franchised restaurants and do not reflect corporate revenue.

McDonald's shares rose 24 cents to close at $94.87 Friday.

On Monday, McNamara may have broken his own record for the largest wave ever surfed. McNamara returned to Nazar?, Portugal, where he set a record a little more than a year ago.

As The Christian Science Monitor reported last May: The Guinness World Records recognized a 44-year-old Hawaii pro surfer for catching a 78-foot wave off the coast of Portugal, saying the November [2011] feat beats a 2008 record for the biggest ridden by more than 1 foot.

Nazare is known as a prime spot for big rides.? The waves roll in off the Atlantic Ocean and travel along an undersea canyon that generate some of the biggest waves on the planet. The Nazare canyon is about 16,000 feet deep in places, and about 140?miles long.

On Monday, :"The conditions in Nazar? were heavenly perfect. Light southern winds and strong swell coming from northwest and hitting the local canyon as it should," according to SurferToday.com.

McNamara is waiting confirmation of the size of the latest wave and whether a new record was set.

McNamara, who began surfing at age 11 and went pro at 17, said the achievement became more important to him when he realized it could help him urge more people to follow their passions.

"The world would be a much better place if everyone was doing what they wanted to do," he told the Associated Press last year after he set the record.
